Epitope mapping of the major capsid protein of type 2 porcine circovirus (PCV2) by using chimeric PCV1 and PCV2.

The immunization of mice with PCV2 virions and the subsequent fusion of their splenocytes with myeloma cells allowed to obtain seven hybridoma which each generated monoclonal antibodies (MAbs) to PCV2 major capsid protein. Each of the seven MAbs was characterized and was shown to be able to recognize a linear epitope of PCV2 capsid protein. The epitope mapping was carried out with PCV1/PCV2-ORF2 chimeras designed by serial deletions of PCV2-ORF2 and replacement with the corresponding sequences of PCV1-ORF2. The seven MAbs used in this study were shown to recognize at least five different but overlapping conformational epitopes. These results contribute to a better understanding of molecular biology and structure of PCV2 capsid protein and virus-cell interaction.
