Brachyspira hyodysenteriae

Protection of pigs from swine dysentery by vaccination with recombinant BmpB, a 29.7kDa outer-membrane lipoprotein of Brachyspira hyodysenteriae.

Escherichia coli was used to produce BmpB, an outer-membrane lipoprotein of Brachyspira hyodysenteriae as a histidine-tagged protein (His6-BmpB) or fused to a maltose-binding protein (MBP-BmpB). Two controlled experiments were carried out to evaluate the level of protection provided by each immunogenic factor. In the first experiment, 20 weaner pigs received His6-BmpB either intramuscularly and orally (10 pigs im/oral) or twice intramuscularly (10 pigs im/im) and compared to unvaccinated controls. In the second experiment, 24 pigs were inoculated intramuscularly with His6-BmpB (12 pigs) or MBP-BmpB (12 pigs) and subsequently given the same protein orally. Two weeks after the oral booster all pigs (included controls) were challenged with Brachyspira hyodysenteriae. MBP-BmpB clearly failed to provide any protection against B. hyodysenteriae. The results obtained with the His6-BmpB were a bit more encouraging but the protein provided only a limited level of protection. In experiment 1 indeed, three pigs vaccinated im/oral and 4 pigs vaccinated im/im developed diarrhea while in the control group not all pigs did (7 pigs out 10 with diarrhea). However, vaccination with His6-BmpB was shown to reduce significantly the incidence of swine dysentery.
